Installation
Set up your Reshape Imaging System by following these steps:
1. Open the cardboard box. If possible keep the box and packaging content.
2. Remove the external top foam.
3. Take out robot and place it on a secure and plain surface.
4. Open the door and remove the foam securing the camera from inside the robot by following the next steps:
- Remove the bottom black foam by pulling the strip.
- Remove the yellow foam.
5. Along with the robot one Petri and one SBS plastic trays are provided. These are used to hold petri- or microtiter plates during imaging.
6. Plug in the provided power cable and ethernet (if you are not planning to run wifi) on the back of the robot.
7. The logo in front should light up after approximately 30 seconds. Understand what the colors on the RIS mean here.
Connect to your Reshape Imaging System
You have received an onboarding email which contains login information. Using this, please log in at discovery.reshapebiotech.com. To test whether the robot has internet connection, use the “New Job” button in the menu (see the figure below). Name the job, select the robot you wish to test, and select at least one of the light presets available. Press “Submit”.
After that, you can go to the job page where the results for this specific job are displayed. Click on "Job List" in the sidebar or refresh the page to update the status of the job. The status should change from "pending" to "started" just a few seconds after the job has been submitted. If this happens, the connection is established. If the status remains "pending," something is most likely blocking the robot from getting an internet connection.
Starting a New Job
To prepare a job, follow these steps:
- Load the plates onto a tray. The lids should always remain on the plates during imaging. This prevents both the drying out of the samples and potential cross-contamination.
- Open the door, slide the tray into the machine, and close the door.
- Log into the user interface at discovery.reshapebiotech.com and start a new job (see the figure below).
Before submitting the job, configure the following:
- Job Name – This can be anything and is only used to differentiate between various jobs.
- Plate Type – Select the type of plate used here. This adjusts the zoom and focusing to match the respective plate type and the method of scanning over the plates for imaging individual wells.
- Plate Selection – Here you can select which plates should be imaged during the job. Dark blue indicates plates to be imaged, while gray indicates plates that will not be imaged. All plate positions are selected by default.
- Light Settings – Choose the types of light to be used during the job. Simply activate one or more presets by toggling the buttons. Generally, the default option (Medium Bottom Light) is suitable for most types of experiments.
- Capture Interval and Duration – Choose whether you want to capture a single image or, alternatively, how often imaging should occur between each capture. If an interval is chosen, you can also set the total duration of image capture and the end date of the job.
- Select the Temperature Control Checkbox – If the job is to be incubated, set the incubation temperature.
Press "Submit". The job status will change to "Started" and imaging will begin within 30-60 seconds.
